Description

To determine the clinical and epidemiological characteristics, as well as evaluate some immunological, immunogenetic and microbiological factors, and their relationship with the vaginal microbiota in a cohort of Colombian patients suffering from recurrent vulvovaginal candidiasis (RVCV).

Specific objectives

1. Describe the main clinical and epidemiological characteristics in a cohort of Colombian patients diagnosed with CVVR.

2. Study the composition of the vaginal microbiota in patients diagnosed with CVVR and the respective control group using metagenomics techniques.

3. Characterize Candida isolates: species identification, susceptibility tests to different antifungals, and biofilm formation.

4. Determine the in situ expression of genes associated with the immune response in patients with CVVR: cytokines (IL-1b, IL-8, IL-10, IL-17, IL-18, TNF-a and TGF-b); specific transcription factors for Th1 (T-bet), Th2 (GATA3), Th17 (RORgt) and Treg (FoxP3) profiles, and enzymes related to oxidative/microbicidal mechanisms: inducible nitric oxide synthase (iNOS), NADPH oxidase 2 ( NOX2) and indolamine deaminase (IDO).

5. To evaluate the different cell populations in peripheral blood (Neutrophils, LT CD4, LT CD8 and NK cells) of patients diagnosed with CVVR.

6. Determine the proliferative capacity of mononuclear cells from patients with CVVR in response to Candida spp antigens.

7. Determine the fungicidal capacity of neutrophils isolated from patients with CVVR against Candida spp yeasts.

8. Determine the serum levels of Dectin-1 and soluble MLB as well as the presence of polymorphisms in the genes that code for Dectin-1 and mannose-binding lectin (MLB) in patients with CVVR.
