Abstract
Assessing the risk of breast cancer (BC) in a woman is essential when advancing towards personalized detection and reduction of morbidity and mortality rates. Mammographic density (MD) is a well-known risk factor and has the potential to improve the quality of risk prediction models, however discriminatory accuracy remains limited at the individual level. Serum metabolic differences according to the percentage of mammographic density could represent an innovative and useful risk identification tool in clinical practice. Objectives: In this pilot study, the possible association and predictive capacity of the serum metabolic profiles of low and high risk of developing breast cancer was evaluated in a group of Colombian women. Sixty patients were recruited from the mastology unit of the Clínica Colombia in Bogotá. Results: A pilot metabolomics approach was used to obtain a global picture of the metabolic alterations that occur at different levels of BC risk by mammographic density. The samples were analyzed by gas chromatography coupled mass spectrometry (GC-MS). Differences between group profiles were assessed with univariate (UVA) and multivariate (MVA) analysis using a nonparametric t-test, principal component analysis (PCA), and orthogonal partial least squares regression (OPLS-DA).
